Giris ve amaç: Mide mukozasinda aktif hidrojen iyonlarinin lokalizas yonunu görüntüleme imkani elde ettikten sonra pariyetal hücrelerin proton pompasini tam blokaja ugrattik ve histokimyasal yöntem kullanarak bu hücrelerin en siki yerlestigi fundus mukozasinda asid sekresyonunu takip ettik. Gereç ve yöntem: Kilosu ve yasi ayni olan köpeklere iki haftali k bir sürede 4, 8 ve 12mg/kg dozda omeprazol verdik. Kontrol amaciyla kullanilan köpege ilaç vermedik ve ayni kosullarda besledik. Sonra hayvanlarin midesinin fundus, korpus ve antrumundan parçalar alarak histokimyasal yöntemle inceledik. Bulgular: Kontrol hayvanin mide mukozasinin her yerinde ve mukozanin tüm katlarinda aktif hidrojen iyonlarinin oldugunu tasbit ettik. 4mg/kg dozdaki omeprazolun etkisinden bu iyonlarin konsantrasyonun azalmasini, 8mg/kg dozun etkisinden iyonlarin kesin azalmasini, 12mg/kg dozdaki ilacin etkisinden ise proton pompasinin tam blokaja ugramis oldugunu, yani boyanmanin olmadigini izledik. Proton pompasi tam blokaja ugratilmis hayvanin mide fundusundan alinmis mukoza örneklerinin bir kismi ise kuru buz üzerinde dondurularak sonradan çözütülmüstü. Bu numunelerde mukozanin yüzeye yakin bölgelerinde boyanma olmadigi halde, mukozanin daha derin katlarinda renglenme izlendi, yani aktif hidrojen iyonlarinin diffüz sekilde ortaya çiktigi görüldü. Sonuç: Mide fundus mukozasinda pariyetal hücreler mukozanin daha derin katlarinda yaygin sekilde yerlesmisler. Burada salgilanan aktif hidrojen iyonlari mukozaya asagidan yukariya, yani lümene dogru, hem de diffüz sekilde yayilmaktadi rlar.
Background/aim: With imaging of active hydrogen ions in stomach mucous membrane we have blocked parietal cells proton pump and observed acid secretion in fundic mucous membrane. Materials and methods: Taking into consideration age and weight, dogs were given omeprazole in 4, 8 and 12 mg/kg dosages and were kept in the same conditions with control dogs. After 14 days, biopsies were taken from fundus, corpus and antrum and investigated by histochemical method. Results: In control dogs, the active hydrogen ions were present in entire specimens of gastric mucous membrane. In dogs administered a 4 mg/kg dosage of omeprazole, a decrease in the number of these ions was observed; in those administered an 8 mg/kg dosage, a further significant decrease was observed, and in those given a 12 mg/kg dosage, the proton pump was fully inhibited and no histochemical staining was seen. The specimens removed from the animals in which the proton pump was fully blocked were frozen twice and melted and also investigated. Diffuse staining was observed on those specimens where parietal cells were placed, but no staining was observed in the surface of the mucous which looks towards the gastric cavity. Conclusion: Parietal cells are located deep in stomach fundic mucous membrane. The transition of active hydrogen ions to the mucous membrane occurs from low side to upside by diffusion.
